慢性间歇低氧对幼鼠学习记忆影响

【摘要 】 目的 建立慢性间歇低氧幼鼠模型 ,研究其空间学习记忆能力和海马 CA1区神经元超微结
构的变化。方法 八臂迷宫训练成功的 3~4周龄 S D幼鼠 40只随机分 2周间歇低氧组 ( 2H)、 对照组
(2C)和 4周间歇低氧组 (4H)、 对照组 ( 4C) ,建立慢性间歇低氧动物模型 ,间歇低氧结束后八臂迷宫测试
学习记忆能力 ,透射电镜观察海马 CA1区神经元的超微结构变化。结果 间歇低氧组参考记忆错误
[ (3 . 30 ± 1 . 06)次、 (2 . 00 ± 0 . 67)次 ]、 工作记忆错误 [ (2 . 30 ± 0 . 67)次、( 1 . 00 ± 0 . 82)次 ]和总错误次数
[ (5 . 40 ± 1 . 78)次、 (3 . 00 ± 0 . 82)次 ]均高于对照组 ( P < 0 . 05) ,且随着间歇低氧时间的延长 ,这种错误次
数增多 ( P < 0 . 05) ;电镜下间歇低氧组海马 CA1区神经细胞的形态、 胞浆内的细胞器均有明显变化。结论
慢性间歇低氧可引起幼鼠空间学习记忆能力下降 ,这可能和海马 CA1区神经元超微结构改变有关。

【Abstract】 Objecti ve To establish a model of inter mittent hypoxia in weanling rats and study the change
of spatial learning and memory abilities and ultrastructure of hi ppocampal CAl neur ons . Methods Forty 32 week2
old~42 week2 old Sp rague Dawley rats which have been trained t o comp lete the 82 Arm (42 ar m baited) radialmaze
were randomly divided int o 22 week2 hypoxia gr oup ( 2H) , 42 week2 hypoxia group ( 4H) , 22 week2 contr ol gr oup ( 2C)
and 42 week2 control group (4C) . I nter mittent hypoxia modelwas induced by an inter mittent hypoxia cabin . At the
end of study, 82 Arm (42 ar m baited) radialmaze was used t o measure spatialmemory in these weanling rats and the
ultrastructure of hi ppocampal CAl neur onswas studied by electr on micr oscope . Results The nadir of ambient oxy2
gen concentrati on was about 7 . 8% and the normal oxygen concentrati on was about 21 . 0%. Both 4H and 2H rats
had significantlymore err ors choices than 4C and 2C rats( P < 0 . 05, respectively) . More RME,WME and TE were
found in 4H gr oup (3 . 30 ± 1 . 06, 2 . 30 ± 0 . 67, 5 . 40 ± 1 . 78) than in 2H gr oup (2 . 00 ± 0 . 67, 1 . 00 ± 0 . 82, 3 . 00
± 0 . 82) ( P < 0 . 05) . The nuclearmembrane and mit ochondria of hi ppocampal CAl neur ons in inter mittent hypox2
ia group s were dist orted compared with those in control gr oup s . Conclusi on Chronic inter mittent hypoxia could
result in damage and lesi on of spatial learning and memory in weanling rats . Itmay be correlated with the changes
of hi ppocampal CA1 neurons .
